چکیده

Alopecia areata (AA) is a common hair loss condition characterized by the acute onset of scarring loss. Recently, JAK inhibitors have been used to treat patients with severe AA who do not respond conventional treatments such as glucocorticosteroids, cyclosporine, and diphenylcyclopropenone. Our previous studies showed an alternative effect on follicle cells, decreasing mitochondrial damage inflammasome activation, thereby improving AA's inflammatory status. However, no reports investigated role in direct growth promotion cells. Therefore, this study β-catenin activation human primary outer root sheath (ORS) IFNγ-treatment decreased increased Dickkopf-related protein 1 (DKK1) expression ORS cells dose-dependent manner, was both cytosol nuclear fraction. Next, we each selective inhibitor, itacitinib, CEP-33779, ritlecitinib, examine which inhibitor regulates β-catenin. Itacitinib CEP-33779 nucleus inhibited IFNγ treatment ritlecitinib slightly reversed compared other inhibitors. Moreover, itacitinib transcriptional manner. shows that JAK1 JAK2 increase activity decrease DKK1 levels, providing additional therapeutic mechanism than inhibiting JAK-STAT pathway gives more information dermatologists use AA.

Sox9 Increases the Proliferation and Colony-forming Activity of Outer Root Sheath Cells Cultured In Vitro.

BACKGROUND β-catenin plays a pivotal role in hair follicle development and hair growth cycle. OBJECTIVE The aim of this study was to identify β-catenin-regulated genes in cultured human hair outer root sheath (ORS) cells. METHODS Primary cultured ORS cells were transduced with recombinant adenovirus expressing N-terminal truncated β-catenin (constitutive active form), and β-catenin-regulate...

Differentiation of nestin-negative human hair follicle outer root sheath cells into neurons in vitro

A specialized quiescent population of hair follicle stem cells, residing in the hair follicle outer root sheath cells (ORSCs), has previously demonstrated pluripotency for differentiation into neural stem cells (NSCs). A previous study indicated that nestin‑positive hair follicle ORSCs are able to differentiate into neurons. However, little has been reported on the isolation of nestin‑negative ...

Manipulation of outer root sheath cell survival perturbs the hair-growth cycle.

Transgenic mice that overexpress the anti-apoptotic gene bcl-xL under the control of the keratin 14 promoter have significantly shorter hair than non-transgenic littermates. The deficit in hair length correlated with a decrease in the duration of anagen, the growth phase of the hair cycle. A prolongation in telogen, the resting phase of the hair cycle, was also observed in adult animals. In the...

Baicalin increases hair follicle development by increasing canonical Wnt/β-catenin signaling and activating dermal papillar cells in mice

Baicalin is a traditional Chinese herbal medicine commonly used for hair loss, the precise molecular mechanism of which is unknown. In the present study, the mechanism of baicalin was investigated via the topical application of baicalin to reconstituted hair follicles on mice dorsa and evaluating the effect on canonical Wnt/β‑catenin signaling in the hair follicles and the activity of dermal pa...

Roles of phosphatidylinositol 3-kinase in root hair growth.

The root hair is a model system for understanding plant cell tip growth. As phosphatidylinositol 3-phosphate [PtdIns(3)P] has been shown in other plant cell types to regulate factors that affect root hair growth, including reactive oxygen species (ROS) levels, cytoskeleton, and endosomal movement, we hypothesized that PtdIns(3)P is also important for root hair elongation. The enzyme that genera...
